T4 RNA Ligase 2 Mutant-01 is a modified double-stranded RNA ligase mutant derived from T4 RNA Ligase 2 (abbreviated as T4 Rnl2) through targeted mutagenesis via enzyme engineering. This enzyme is also derived from the T4 bacteriophage and belongs to the class of ATP-dependent double-stranded RNA ligases (dsRNA ligases). It is primarily used for ligation reactions at nick sites within double-stranded RNA (dsRNA) structures.
Compared to wild-type T4 Rnl2, the T4 RNA Ligase 2 Mutant-01 demonstrates higher ligation activity and substrate compatibility on 2′-modified nucleotides and phosphorothioate (PS) modified substrates. It is particularly suitable for the ligation of highly modified or fully modified oligonucleotides, such as siRNA and ASO, and offers superior applicability in enzymatic synthesis and large-scale applications.
